Penicillin‐binding protein 3 (PBP3) of Pseudomonas aeruginosa is a membrane bound transpeptidase located on the periplasmic side of the inner membrane. It is involved in the synthesis of the peptidoglycan component of the bacterial cell wall by catalysing the cross‐linking of ᴅ‐alanine to form N ‐acetylglucosamine‐ N ‐acetyl muramic polymers [ 1 ] .

Penicillin-binding proteins (PBPs) function in the late steps of murein biosynthesis (Probable). Probably required for both cortical and vegetative peptidoglycan synthesis (Probable). Although not usually required for cell division, in the absence of PBP 2B (pbpB) it becomes essential. Confers resistance to oxacillin and cephalexin (PubMed:28792086).

PBP3 is a class C PBP with dd -carboxypeptidase (CPase) activity; it trims pentapeptides to tetrapeptides [24]. The ld -CPase LdcB (also called DacB) trims tetrapeptides to tripeptides, which are abundant in PG, and is not a member of the PBP family of proteins [25]. In bacteria, biogenesis of cell wall at the division site requires penicillin‐binding protein 3 (PBP3) (or FtsI).

Penicillin-Binding Protein 3 Is Essential for Growth of Pseudomonas aeruginosa. Chen W(1), Zhang YM(1), Davies C(2). Author information: (1)Department of Biochemistry and Molecular Biology, Medical University of South Carolina, Charleston, South Carolina, USA.

PRINCIPLES OF We have solved two crystal structures of penicillin-binding protein (PBP) 3 (PBP3) from MRSA, the apo form and a complex with the β-lactam antibiotic cefotaxime, and used electrospray mass spectrometry to measure its sensitivity to a variety of penicillin derivatives The soluble form of penicillin-binding protein 3 (sPBP3 ∗) from Streptococcus pneumoniae was expressed in Escherichia coli as a six-histidine fusion protein. The protein was purified and used to develop a microplate assay in direct competitive format for the detection of penicillins and cephalosporins in milk. Penicillin-binding protein (PBP) 3, or ftsI, is an essential transpeptidase in Mycobacterium tuberculosis (Mtb) required for cell division, and thus it is an important drug target. Structures of apo Mtb PBP3 and of complexes with five β -lactams, including meropenem and faropenem, reveal how they cause inactivation via formation of hydrolytically stable acyl-enzyme complexes.
